QIAGEN Buffer AL Lysis LOT Q(4)
 
QIAGEN Buffer AL is a specialized cell lysis and binding buffer essential for genomic DNA extraction and purification using the manufacturer's flagship spin-column kits. It plays a dual role by breaking open cellular membranes to release nucleic acids and establishing the precise chemical environment required for DNA to bind to a silica membrane.
Key Characteristics & Applications
  • Core Mechanisms: It contains a high concentration of guanidine hydrochloride (a chaotropic agent) and malic acid. This denatures cellular proteins, deactivates nucleases, and alters the hydration structure of nucleic acids to facilitate spin-column binding. 
  • Primary Compatibility: It is supplied ready-for-use and explicitly formulated for the QIAGEN QIAamp and QIAGEN DNeasy extraction protocols.
  • Storage Artifacts: It is highly stable for 1 year at room temperature (15–25°C). However, it is prone to forming a crystalline precipitate during storage. If this happens, you must warm the buffer bottle to 55°C until the crystals completely dissolve before starting your protocol.
Usage Differences: Blood vs. Tissue Protocols
A common point of confusion in the lab is why some workflows require an additional buffer. The requirement changes depending on the mechanical complexity of your biological starting material: 
  • Blood Protocols: Use Buffer AL alone (often alongside a protease) because blood cells are highly accessible and lyse easily.
  • Tissue Protocols: Require an initial digestion step using Buffer ATL (an SDS-based tissue lysis buffer) and Proteinase K to break down tough extracellular matrices. Afterward, Buffer AL must still be added to the digested mix to create the necessary conditions for the DNA to bind to the silica column membrane.
Reagent Specifications
  • Buffer AL (264 mL): This standalone volume provides a reliable reserve for scaling manual extractions or offsetting automated reagent dead volumes.
    • Justification: Sourced as an individual reagent bottleneck solution for workflows processing high-throughput batches across QIAGEN Kits. 
Special Preparation for DNeasy 96 Kits
When adjusting workflows for high-throughput multi-well plates, you must manually modify the reagent: 
  1. If using the DNeasy 96 Blood & Tissue Kit, you must blend specific ratios of 96–100% ethanol directly into Buffer AL to create a working solution termed Buffer AL/E. 
  2. For example, a standard preparation mix requires 100 mL of Buffer AL blended with 108 mL of pure ethanol. Once mixed, this solution remains chemically stable for 3 months at room temperature

LIMITED PICKUP DATES/TIMES:
Currently, pickup dates and times are EXTREMELY LIMITED. Pickups will only be available to be scheduled on Thursdays of each week, and within a timeframe of 10:00am and 1:00pm. Scheduling a specific date/time is STILL REQUIRED! This notice supersedes all other pickup times given elsewhere in the auction information.

The photos below are example images, and the item you win may be slightly different than what is shown. The Pickup Location may be different than the one shown in the auction. The winner will be given the correct address once payment has been given.


**Notice to Bidders**
WE DO NOT SHIP ITEMS.
The winning bidder is responsible for supplying proper manpower, equipment (i.e. dolly, handtruck, etc.), and appropriate vehicles for pickup. No equipment or manpower will be supplied by the County of Sonoma. You are required to pick-up all items included in the auction.

Winning bidder must make an appointment, and confirm it(!), with the auction contact to arrange pick-up date/time. DO NOT come to the pickup location without an appointment! You will not receive your items if you do!

Please note that pickup time/date availability will vary, however, pickups are usually able to be scheduled on Thursdays, at a scheduled time between 10am and 1pm. Due to specific item locations and Auction Contact availability, exceptions may be made. If in doubt, please contact the Auction Contact for more information.

We do not ship items, so we do not accept any pre-paid shipping labels. If you are unable to pick up your auction winnings in person, you may specify a family member or friend to pick up the item for you, or, you may hire a courier/freight company to pick up, pack, and ship the item to you. If you require a courier/freight company to pick up your auction winnings, please note that 1) you must reach out to the Auction Contact and inform them of this before doing any pickup scheduling, and 2) we are unable to put items in boxes, or secure them to pallets, so the courier/freight company you chose must be able to do that themselves if needed.

Payment must be received within 5 calendar days of auction ending, or else the item(s) may be relisted. If the item(s) are not picked up within 3 weeks of the auction ending, the item(s) may be relisted, and no refunds will be given.

It is the responsibility of the Bidder to fully understand what it is you are bidding on. Some items can be extremely heavy, require a specific type of vehicle to transport, or take considerable time to remove. The County shall deny access to pick up auction winnings if the bidder arrives unprepared to SAFELY and expediently remove said property from the premises within the time allocated for the pickup, regardless of distance traveled or under any other circumstance.

If you cannot safely remove the items within the time allocated you will be asked to leave the premises. NO EXCEPTIONS WILL BE GRANTED, and NO REFUND provided. Do not arrive by yourself to pick up large or heavy items that require assistance. If you cannot safely load items purchased in the allocated time, you will be asked to leave the premises and either forfeit the transaction or arrange to come back within two business days for pickup if you still have pickup time available.
