QIAGEN Protease and QIAGEN Protease Solvent LOT Q(17)

QIAGEN Protease and QIAGEN Protease Solvent are specialized reagents used together to degrade proteins, nucleases, and contaminants during DNA and RNA isolation workflows.
System Overview
  • QIAGEN Protease: A recombinant serine endopeptidase derived from Bacillus. It is supplied as a lyophilized (freeze-dried) powder to maximize long-term stability at room temperature.
  • QIAGEN Protease Solvent: A specialized aqueous solution formulated with 0.04% sodium azide as a preservative. It is specifically used to reconstitute the lyophilized protease powder into a ready-to-use liquid enzyme.
Reconstitution Instructions
The volume of solvent or water required depends heavily on the specific kit protocol being used. Always check your kit manual before mixing: [1, 2, 3]
  • QIAamp DNA Blood (Manual) Kits: Reconstitute one vial of protease with 7.0 mL of solvent or distilled water.
  • QIAamp BioRobot Kits: Reconstitute one vial of protease with 10.0 mL of distilled water. [1]
Storage and Stability
  • Lyophilized Powder: Stable for up to 12 months when stored dry at room temperature (15–25°C). For longer shelf life, store at 2–8°C.
  • Reconstituted Liquid: Stable for up to 2 months when stored at 2–8°C.
  • Long-term Storage: Can be stored at –20°C for up to 6 months. It is highly recommended to divide the solution into aliquots before freezing to prevent activity loss from repeated freeze-thaw cycles.
Performance Profile
  • Optimal Conditions: The enzyme exhibits broad substrate specificity and remains highly active in alkaline conditions (pH 7.5–10.5) and elevated temperatures up to 55°C.
  • Denaturant Compatibility: Its activity actually increases in the presence of common lysis buffers containing denaturants like urea, guanidine HCl, or up to 1% SDS.
  • Inhibitor Resistance: It is not inhibited by up to 100 mM EDTA in standard buffers, allowing it to function while nucleases are safely deactivated.
  • Inactivation: Easily inactivated by heating the reaction mixture to 70°C for 15 minutes.
Comparison: QIAGEN Protease vs. Proteinase K
While both serve similar primary goals, they feature crucial differences:
  • Target Residues: Protease cleaves preferentially at neutral and acidic amino acid residues, whereas Proteinase K targets the carboxyl side of hydrophobic, aliphatic, and aromatic residues.
  • Digest Speed: Proteinase K is preferred for highly complex tissues requiring rapid digestion times.
  • Cost: QIAGEN Protease serves as a highly economical alternative to Proteinase K for isolation from straightforward samples like liquid blood and cultured cells.
